Purpose

This online database is dedicated to documenting staphylococcal protein A (spa) sequences in Staphylococcus pseudintermedius and was created to quickly identify and catalog new tandem repeat sequences and spa types. Please note that the spa types for S. pseudintermedius and S. aureus are different, and the database for S. aureus can be found at the Ridom Spa Server. If you have a tandem repeat sequence or spa type not in this database or were previously assigned a number, please use the contact form so the database can be updated. Dr. Arshnee Moodley is the curator of the spa database and any new repeat sequences or types (including forward and reverse .ab1 and .seq files) should be submitted to her to be assigned a number.

Spa-typing method

Spa-typing in S. pseudintermedius was first described by Moodley et al. (2009) using the primer sequences: SIspa F (5'-AACCTGCGCCAAGTTTCGATGAAG-3') and SIspaR (5'-CGTGGTTTGCTTTAGCTTCTTGGC-3'). A nested PCR method was developed by Perreten et al. (2010) using the outer primers SPspa1F (5′-CCGCTCTATTTTTAGGTTAATC-3′) and SIspaFlkR1 (5′- CGTAACAACTCAATGCTACATA-3′) to achieve better amplification in isolates that are not able to be typed solely with the Moodley primer set.
